Werner Helicase Inhibitor RO7589831 Enters Phase 1 Testing in MSI-High Tumors
Key Takeaways
- Nature Medicine published a phase 1 trial of RO7589831, a first-in-class Werner helicase inhibitor, in advanced MSI-high solid tumors on 29 September 2026.
- The report covers the dose-escalation stage, focused on safety, tolerability, pharmacokinetics, and dose selection.
- The mechanism rests on synthetic lethality: MSI-high cancer cells depend on WRN function in a way that mismatch repair-proficient cells do not.
- Dose-escalation data are early and exploratory, so no efficacy conclusions can be drawn from this stage alone.
